Allergome-wide peptide microarrays enable epitope deconvolution in allergen-specific immunotherapy

چکیده

BackgroundThe interaction of allergens and allergen-specific IgE initiates the allergic cascade after crosslinking receptors on effector cells. Antibodies other isotypes may modulate such a reaction. Receptor requires binding antibodies to multiple epitopes allergen. Limited information is available complexity epitope structure most allergens.ObjectivesWe sought allow description IgE, IgG4, IgG recognition at global, allergome-wide level during immunotherapy (AIT).MethodsWe generated an microarray comprising 731 in form more than 172,000 overlapping 16-mer peptides. Allergen by was examined serum samples collected from subjects undergoing AIT against pollen allergy.ResultsExtensive induction linear peptide-specific Phl p 1– Bet v 1–specific humoral immunity demonstrated 3-year-long grass birch allergy, respectively. Epitope profiles differed between but were largely established already 1 year AIT, suggesting that dominant antibody clones remained as important contributors following their initial establishment early phase AIT. Complex, subject-specific patterns allergen isoform group cross-reactivities repertoires observed, indicate different levels protection sources.ConclusionsThe study highlights nature recognized We envisage deconvolution will be aspect future efforts describe analyze outcomes personalized manner. Recent Progress in Allergen Immunotherapy

The efficacy of allergen immunotherapy for the treatment of allergic rhinoconjunctivitis with or without seasonal bronchial asthma and anaphylaxis caused by the sting of the hymenoptera class of insects has been clearly demonstrated in numerous well-designed, placebo-controlled trials. Immunotherapy whether by subcutaneous injection of allergen extract or by oral/sublingual routes modifies peri...

Allergen-specific immunotherapy

Allergen-specific immunotherapy is a potentially disease-modifying therapy that is effective for the treatment of allergic rhinitis/conjunctivitis, allergic asthma and stinging insect hypersensitivity. However, despite its proven efficacy in these conditions, it is frequently underutilized in Canada. The decision to proceed with allergen-specific immunotherapy should be made on a case-by-case b...

Allergen Specific Immunotherapy in Asthma

The use of allergen-Specific Immunotherapy (SIT) to treat asthma remains one of the most debated aspects in the field since several decades. The available guidelines provide no clear or unequivocal indication, stating only that SIT is specific for the allergen causing the allergic disease (rhinitis and/or asthma) and not for the disease itself [1]. Nonetheless, in the majority of the clinical t...

Allergen-Specific Immunotherapy in Asthma

Current asthma therapies can effectively control symptoms and the on-going inflammatory process; however, they do not affect the underlying, dysregulated immune response. Thus, they are limited to blunting the progression of the disease, which relapses on ceasing the treatment. Allergen-specific immunotherapy (AIT) is the only etiology-based treatment capable of disease modification. Recent evi...

Allergen-specific immunotherapy in children

HISTORICAL TIPS The science of “Immunology” originated in the 19th century and grass pollens were identified for the first time as the likely trigger of seasonal hay fever in the 1870s. Skin allergy testing became an accepted assessment technique around 1910. Immunoglobulin E (IgE) was identified in the 1960s. The first scholarly report of immunotherapy for allergy appeared by Noon and Freeman ...
